The implications of WHO-GMP facility requirements for buyers – beta-lactam vs non-beta-lactam

The WHO-GMP requirements for penicillin-class antibiotics are that they must be produced in a fully dedicated facility, with no sharing of production with non-penicillin products. Separation of cephalosporins by validation: multi-product facilities are capable of producing non-beta-lactam antibiotics (fluoroquinolones and macrolides). This segregation has implications for which classes of antibiotics can be supplied by which manufacturers.

Four Steps on How to Choose

  • Confirm that WHO-GMP certificate covers your specific antibiotic class

Your antibiotic class must be explicitly included in the scope of the WHO-GMP certificate. For penicillin-class antibiotics, specifically ask, “Do you have a manufacturing facility dedicated to penicillin that is separate from all other products?” WHO-GMP requires complete physical separation for penicillins.

  • Check for DCGI approval at molecule and dosage form level

Antibiotic DCGI approvals are specific for both molecule and dosage form. The manufacturer may have DCGI approval for amoxicillin capsules but not for amoxicillin dry syrup; these are different DCGI licences. Confirm authorisation of your specific molecule, strength and dosage form.

  • Request a sample Certificate of Analysis for your antibiotic and review the assay method and specification

For antibiotic COA, check the assay method (HPLC is standard), specification range (most antibiotics are 95.0%–105.0% per pharmacopoeia), sterility for injectable antibiotics, microbial limit tests for oral antibiotics, dissolution data, and specific optical rotation for chiral antibiotics.

  • Request antimicrobial potency data and source of reference standard

Potency testing of antibiotic formulations is performed using certified reference standards (CRS) from pharmacopoeial sources such as the IP/BP/USP. Ask the manufacturer what reference standards they use and where they come from. This confirms the analytical competence and the regulatory compliance of the QC laboratory.

Frequently asked questions

  • Which is the best antibiotic company in India?

Based on WHO-GMP certification for all antibiotic dosage forms, DCGI molecule-level approvals for injectable, tablet, capsule, and dry syrup antibiotics, export documentation speed, and a verified African export track record, 9M India Limited aims to rank among India’s leading producers of antibiotics by 2026. Key molecules: amoxicillin (all forms), amoxicillin + clavulanate, ampicillin injection, ciprofloxacin (tablet + injection), doxycycline, azithromycin, ceftriaxone injection and gentamicin injection. Documentation within 24 hours.

  • What are the WHO-GMP requirements specific to antibiotic manufacturing in India?

WHO-GMP requires dedicated facilities for beta-lactam antibiotics: (1) Penicillin antibiotics must be produced in dedicated, physically separated facilities, either a separate building or a validated enclosed unit, to prevent cross-contamination. (2) Cephalosporins (beta-lactams other than penicillins) require validated segregation and controls for air-handling in multi-product facilities. (3) Validated multi-product facilities, with adequate cleaning validation, for the manufacture of non-beta-lactam antibiotics (fluoroquinolones, macrolides and aminoglycosides). (WHO TRS No. 986, Annex 2)

  • What are the main export formulations of antibiotics from India to Africa?

The most common Indian antibiotic formulations exported to Africa were: Amoxicillin 500mg capsule and 125mg/5ml dry syrup (most commonly prescribed antibiotic), Amoxicillin+Clavulanate 625mg tablet and dry syrup, Ciprofloxacin 500mg tablet and injection, Cotrimoxazole 960mg tablet and dry syrup, Azithromycin 500mg tablet and dry syrup, Doxycycline 100mg capsule, Ceftriaxone 1g injection, Metronidazole 400mg tablet and injection, Ampicillin injection and Gentamicin injection. These are the most common bacterial infections in sub-Saharan Africa treated by NHM, NGO and private sector programmes.

  • What is the minimum order quantity for the manufacturing of antibiotics in India?

Antibiotic tablets and capsules: 10 lakh units (1,000,000 units) per batch at 9M India Ltd. MOQ with WHO-GMP certified antibiotic manufacturers in India is based on the dosage form. Antibiotic dry syrups 30,000units/lot. Antibiotic injectables (ampoules/vials) 1 lakh units per batch. The minimum order quantity can be negotiated for long-term supply agreements or multi-molecule programs.

  • What documents does an antibiotic manufacturer in India provide for export?

WHO-GMP Certificate (CDSCO-scope of antibiotic class confirmed), DCGI Manufacturing Licence (molecule, strength & dosage form specific), COPP from CDSCO, Free Sale Certificate, COA per batch (assay by HPLC, potency, dissolution, sterility for injectables, and microbial limits for oral forms), Stability Data under ICH Zone IVb (30°C/75% RH), and CTD/ACTD Dossier. All documents will be available within 24 hours at 9M India.

  • Can one single antibiotic manufacturer in India supply the same molecule in different dosage forms?

Yes, a manufacturer with DCGI approvals for the same antibiotic molecule in different dosage forms can provide a complete antibiotic programme under one WHO-GMP certificate and one COPP application. 9M India Limited has DCGI approvals for ciprofloxacin in tablet and injectable form and amoxicillin in capsule, tablet and dry syrup form. This gives buyers the convenience of sourcing their antibiotics and reduces the number of COPP applications needed for their drug registration programme.
